Mucins are a family of macromolecules that line and protect the respiratory epithelium from microbes and pollutants in the local environment. Of the family members that are known to date, some are produced in a cell type and tissue-specific manner, suggesting distinct biological roles for members. Some members polymerize after secretion to form gel-like substances that coat the epithelial layer. MUC16 is a member of the mucin family, which are high molecular weight, O-glycosylated proteins that play an important role in forming mucous barrier and are found on the apical surface of the epithelia. It contains an extracellular domain at its amino acid terminus, a large tandem repeat, and a transmembrane domain with a short cytoplasmic domain. MUC16 has been linked to lung cancer and ovarian cancer. It provides protective, lubricating barrier against particles and infectious agents at mucosal surfaces. Product :

  Immunohistochemical analysis of MUC16 staining in human mesothelioma formalin fixed paraffin embedded tissue section. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.0). The section was then incubated with the antibody at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.

Describe :

Blocking peptides are peptides that bind specifically to the target antibody and block antibody binding. These peptide usually contains the epitope recognized by the antibody. Antibodies bound to the blocking peptide no longer bind to the epitope on the target protein. This mechanism is useful when non-specific binding is an issue, for example, in Western blotting (WB) and Immunohistochemistry (IHC). By comparing the staining from the blocked antibody versus the antibody alone, one can see which staining is specific; Specific binding will be absent from the western blot or IHC performed with the neutralized antibody.
