MHC class II (MHC-II) proteins play critical roles in cellular immune responses and their expression is mainly regulated by the non-DNA binding transcription factor CIITA (MHC class II transactivator) . CIITA expression is upregulated by IFN-γ and it in turn enchances MHC-II expression and represses collagen expression . CIITA has a limited number of transcriptional targets, most of which are involved in MHC-mediated antigen presentation . Mutations in the CIITA are associated with the hereditary immunodeficiency disease Bare Lymphocyte Syndrome (BLS) which is characterized by a nearly complete absence of MHC-II expression (also referred to as MHC-II deficiency). Blocking peptides are peptides that bind specifically to the target antibody and block antibody binding. These peptide usually contains the epitope recognized by the antibody. Antibodies bound to the blocking peptide no longer bind to the epitope on the target protein. This mechanism is useful when non-specific binding is an issue, for example, in Western blotting (WB) and Immunohistochemistry (IHC).
