This gene encodes a tumor suppressor protein containing transcriptional activation, DNA binding, and oligomerization domains. The encoded protein responds to diverse cellular stresses to regulate expression of target genes, thereby inducing cell cycle arrest, apoptosis, senescence, DNA repair, or changes in metabolism. Mutations in this gene are associated with a variety of human cancers, including hereditary cancers such as Li-Fraumeni syndrome. Alternative splicing of this gene and the use of alternate promoters result in multiple transcript variants and isoforms. Additional isoforms have also been shown to result from the use of alternate translation initiation codons from identical transcript variants (PMIDs: 12032546, 20937277). Product :

  Western blot analysis of extracts of HeLa cells, using Phospho-p53-S46 antibody at 1:1000 dilution.HeLa cells were treated by Calyculin A at 37℃ for 30 minutes after serum-starvation overnight.
  Secondary antibody: HRP Goat Anti-Rabbit IgG at 1:10000 dilution.
  Lysates/proteins: 25ug per lane.
  Blocking buffer: 3% nonfat dry milk in TBST.
  Detection: ECL Basic Kit .
  Exposure time: 180s.

  Immunoprecipitation analysis of 200ug extracts of 293T cells, using 3 ug Phospho-p53-S46 pAb . Western blot was performed from the immunoprecipitate using Phospho-p53-S46 pAb at a dilution of 1:1000. 293T cells were treated by UV at room temperature for 30 minutes after serum-starvation overnight, and then treated by 10% FBS at 37℃ for 30 minutes.

Describe :

Blocking peptides are peptides that bind specifically to the target antibody and block antibody binding. These peptide usually contains the epitope recognized by the antibody. Antibodies bound to the blocking peptide no longer bind to the epitope on the target protein. This mechanism is useful when non-specific binding is an issue, for example, in Western blotting (WB) and Immunohistochemistry (IHC). By comparing the staining from the blocked antibody versus the antibody alone, one can see which staining is specific; Specific binding will be absent from the western blot or IHC performed with the neutralized antibody.
