The protein encoded by this gene is phosphorylated by ZAP-70/Syk protein tyrosine kinases following activation of the T-cell antigen receptor (TCR) signal transduction pathway. This transmembrane protein localizes to lipid rafts and acts as a docking site for SH2 domain-containing proteins. Upon phosphorylation, this protein recruits multiple adaptor proteins and downstream signaling molecules into multimolecular signaling complexes located near the site of TCR engagement.   Western blot analysis of extracts of Jurkat cells, using Phospho-LAT-Y171 antibody at 1:1000 dilution. Jurkat cells were treated by Hydrogen Peroxide for 2 minutes after serum-starvation overnight or treated by CD-3 for 2 minutes after serum-starvation overnight.
  Secondary antibody: HRP Goat Anti-Rabbit IgG at 1:10000 dilution.
  Lysates/proteins: 25ug per lane.
  Blocking buffer: 3% BSA.

Describe :

Blocking peptides are peptides that bind specifically to the target antibody and block antibody binding. These peptide usually contains the epitope recognized by the antibody. Antibodies bound to the blocking peptide no longer bind to the epitope on the target protein. This mechanism is useful when non-specific binding is an issue, for example, in Western blotting (WB) and Immunohistochemistry (IHC). By comparing the staining from the blocked antibody versus the antibody alone, one can see which staining is specific; Specific binding will be absent from the western blot or IHC performed with the neutralized antibody.
